First-Class Mail and other mail services are not affected.

Yes, the rates will be automatically updated on any listing that uses Calculated USPS Shipping.
Note that if you sell something on April 25th but don't ship the same day, the buyer will be charged the old rate but the seller will be charged the new rate when purchasing the label.
