Uncooperating Seller and something else
1. Seller doesn't seem to accept my cancellation of the item he's selling.
2. The item was to be picked up.
3. I did not pick it up because I changed my mind on the purchase.
4. When filling out the return form of eBay's, it asks if I sent it by FedEx or USPS.
5. Number 4 does not apply to me because it was a pickup, and I had not even picked it up.
6. My credit card was charged. It should not have been charged until I picked up the item.

Are you sure your credit card was charged? Or was it a PENDING charge? That isn't the same as a charge you are responsible for paying for. It is kind of like a placeholder waiting for the real charge to come through.
You can't file for the Return of something you don't have. So you have been trying to open the wrong type of claim.
If it is an actual charge on your credit card. Go to your Credit Card's website and put the amount in Dispute. Don't use the reason as you changed your mind, tell them you never received the item, which would be correct. Then Ebay will get notified as well as the seller that you filed a Chargeback. It will take a little time to get it all processed and settled, but you will have your money protected.
