Purchased 2 items in error should have been just 1
I purchased 2 bottles of perfume in error, should have bought only 1. I immediately texted seller requesting to cancel order. The seller refuses to cancel the order saying she is charged $23 for canceled orders. Is this true, refunds cost the seller money?

No, it's not true. If the seller cancels, especially as 'buyer requested', all their fees are credited back automatically.
Ask her to explain the $23 charge - maybe we're missing something?
