Yes you can offer local pick up as one of your delivery options or you can have it as the only option. When you list an item it is in the shipping section.
You can not change the delivery method after the sale therefor you can not have a buyer pick up if you did not have it as an option for the buyer to choose when they purchased.
If you have a buyer who wants to pick up an item you have already sold the way to handle it is to cancel the sale using problem with buyer's address and relist the item with the correct shipping method for them to repurchase. With local pick up the buyer receives a QR code and a digit code that the seller enters into the eBay APP as proof of delivery. The page below has information on how local pick up works.

When you add local pick up the buyer can pay on eBay before picking up, you can also accept cash if the buyer wants to pay that way but they still have to purchase the item on eBay just not pay on eBay. You would still need to scan the code for proof of pick up.
